Pyrroloquinoline quinone — PQQ — gets one of the boldest pitches in the energy-supplement aisle: it doesn’t just support your mitochondria, it helps you make new ones. Marketing copy often pairs it with CoQ10 as a “build and fuel” combination, and it commands a premium price for a compound you take 20 milligrams of.
It’s a real molecule with real biochemistry behind it. It’s also a good case study in how a mechanism that looks impressive in a petri dish gets sold as a proven human benefit.
What PQQ Is
PQQ is a small quinone compound first identified in bacteria, where it acts as a redox cofactor — a helper molecule that shuttles electrons in certain enzyme reactions. It’s unusually stable, capable of cycling through thousands of oxidation-reduction reactions without breaking down, which is where a lot of its antioxidant reputation comes from.
In humans, PQQ is not an established essential nutrient. Researchers proposed at one point that it might qualify as a new vitamin, and that framing still circulates in supplement copy, but the claim was contested and no human PQQ-dependent enzyme has been confirmed. There is no RDA, no recognized deficiency state, and no diagnostic test for PQQ status.
It does show up in food, in trace amounts. Fermented soybeans (natto) are the standout, with parsley, green peppers, kiwifruit, green tea, and human breast milk also containing measurable quantities. We’re talking micrograms — dietary intake is usually estimated in the range of tens of micrograms a day, while a capsule delivers 10,000 to 20,000 micrograms. That’s a pharmacological dose, not a dietary one, and it’s a reason to be careful about extrapolating from “it’s found in food” to “it must be safe at any dose.”
Where the Mitochondria Claim Comes From
The biogenesis story traces back to laboratory work showing that PQQ influences signaling pathways involved in mitochondrial production — most often cited is activation of PGC-1α, a regulator that helps switch on the genes involved in building new mitochondria. Rodent studies have reported increases in mitochondrial content and markers of mitochondrial function, and animals raised on PQQ-deficient diets showed reduced mitochondrial numbers alongside growth and reproductive problems.
That’s genuinely interesting biology. What it isn’t is evidence that a 20 mg capsule increases mitochondrial density in a human being. Nobody has taken muscle biopsies from supplemented people and counted mitochondria in a well-powered trial. The mechanism is plausible and the endpoint is unmeasured — which is a very common gap, and one worth spotting whenever a product markets a mechanism rather than an outcome. What Human Studies Have Found
The human literature on PQQ is small. A handful of controlled trials, mostly conducted in Japan, have used 20 mg/day for 8 to 12 weeks in groups of roughly 17 to 40 adults:
- Fatigue and sleep. Studies in adults reporting fatigue or poor sleep have described improvements in self-reported sleep quality, sleep duration, and fatigue scores relative to baseline or placebo. These are subjective outcomes in small samples, exactly where placebo effects run strongest.
- Oxidative stress and inflammatory markers. Short trials have reported changes in blood and urinary markers of oxidative stress after PQQ supplementation. Marker changes are hypothesis-generating; they don’t demonstrate that anyone feels or functions better.
- Metabolic signatures. One small study analyzing urinary metabolites after single doses reported shifts consistent with altered mitochondrial-related metabolism. Suggestive, but a long way from a clinical endpoint.
A few trials have combined PQQ with CoQ10, reporting cognitive test improvements in older adults. Combination designs make it impossible to attribute anything to PQQ specifically — CoQ10 has its own body of evidence, and the pairing is at least as much a marketing convention as a scientific one.
None of this makes PQQ useless. It makes it unproven, which is a different and more accurate word.
Dosing and Practical Notes
- Common dose: 10-20 mg/day. The 20 mg figure is where most of the human data sits, and there’s no evidence supporting the higher-dose products that occasionally appear.
- Form: most supplements use the disodium salt (BioPQQ and similar branded ingredients are the common raw materials). Label weights sometimes refer to the salt rather than PQQ itself, so a “20 mg” product may deliver slightly less active compound.
- Timing: PQQ is water-soluble and doesn’t require fat for absorption. Morning dosing is a reasonable default — a subset of users report it feels stimulating and interferes with sleep taken late, though the sleep studies dosed it in the morning and reported the opposite effect. Individual response varies more than the marketing admits.
- Trial period: if you’re testing it, give it 8-12 weeks to match study duration, and track one or two specific things rather than a vague sense of energy.
If persistent fatigue is your actual concern, PQQ is a poor first move. Safety
Short-term safety at 10-20 mg/day looks reasonable in the trials conducted so far, with no consistent pattern of serious adverse events. The caveats:
- Reported side effects: headache, drowsiness or, conversely, difficulty sleeping, and occasional GI discomfort. Most reports come from higher doses or from combination products.
- No established upper limit. High-dose animal work has shown kidney effects at doses vastly beyond human supplement levels, but the absence of a defined human upper limit reflects sparse data, not proven safety.
- Long-term use is unstudied. The longest human trials run a few months. Nobody knows what daily use for years does.
- Pregnancy and nursing: no adequate data. Don’t use it.
- Medications: no well-documented interactions, largely because so little research exists. If you take medication for a chronic condition, mention it to your prescriber rather than assuming a clean slate.
- Quality: PQQ is expensive to manufacture, which makes it a target for underdosing and adulteration. Prefer products using a named, third-party-tested raw ingredient.
Bottom Line
PQQ is a legitimate research compound sold on a claim its human evidence doesn’t yet support. The mitochondrial-biogenesis story is built on cell and animal work; the human trials are a small handful of short studies at 20 mg/day reporting mostly subjective or marker-level changes. If you want to experiment, 20 mg/day for 8-12 weeks is the researched approach — just go in knowing you’re an early adopter, not a beneficiary of settled science, and spend your first supplement dollars on the boring things with better data.
